In the canonical salvage pathway, nicotinamide phosphoribosyltransferase, known as NAMPT, transfers a phosphoribosyl group to nicotinamide and releases NMN. A second enzyme, NMN adenylyltransferase, then attaches an adenylyl group to NMN to form NAD+. Alternative routes exist, including a pathway that uses nicotinamide riboside and its phosphorylated forms. The relative contribution of extracellular NMN to intracellular NAD+ pools remains an area of active investigation, and the roles of specific transporters and enzymes are not completely defined.
NMN is present in small amounts in various foods, including certain vegetables, fruits, and milk, though dietary quantities are generally low. Laboratory research often uses synthetic or enzymatically produced NMN. The compound has drawn interest because NAD+ levels decline with age in some tissues and because restoring NAD+ may affect metabolism in animal models. Whether oral NMN produces meaningful NAD+ increases in humans and whether such changes translate into health benefits are not fully established.
Nicotinamide mononucleotide, abbreviated NMN, is a naturally occurring nucleotide found in cells. Its structure combines a nicotinamide ring, a ribose sugar, and a phosphate group. As an intermediate in the NAD+ salvage pathway, NMN is converted to nicotinamide adenine dinucleotide, a coenzyme central to cellular redox reactions. NAD+ also serves as a substrate for enzymes involved in DNA repair, stress responses, and metabolic regulation. The compound is therefore part of normal cellular biochemistry rather than an exclusively synthetic molecule.
Two enzymatic steps define the canonical route from nicotinamide to NAD+. Nicotinamide phosphoribosyltransferase, known as NAMPT, produces NMN from nicotinamide and phosphoribosyl pyrophosphate. NMN adenylyltransferases, or NMNAT enzymes, then couple NMN with ATP to form NAD+. Whether intact NMN crosses cell membranes efficiently remains an active area of investigation; some studies propose direct transport, while others emphasize extracellular dephosphorylation to nicotinamide riboside followed by uptake. The relative contribution of each route likely depends on cell type, tissue, and experimental conditions.

Nicotinamide mononucleotide, abbreviated NMN, is a nucleotide composed of nicotinamide, ribose, and phosphate. Its structure links nicotinamide to D-ribose 5-phosphate through a glycosidic bond, placing it in the pyridine nucleotide family. The compound exists in alpha and beta anomeric forms, and the beta form is the one used in NAD+ biosynthesis. NMN is not a protein or a hormone; it is a small water-soluble molecule that occurs in living cells as a metabolic intermediate.
Natural sources of NMN include mammals, plants, and microorganisms, where it functions as an intermediate in NAD+ salvage and biosynthesis pathways. In mammals, the enzyme nicotinamide phosphoribosyltransferase produces NMN from nicotinamide and phosphoribosyl pyrophosphate. NMN is then converted to NAD+ by nicotinamide mononucleotide adenylyltransferase. Some foods contain measurable NMN, but reported amounts vary widely by species, tissue, and analytical method. The extent to which dietary NMN contributes to cellular NAD+ pools remains an open research question.
Chemically, NMN is described by the molecular formula C11H15N2O8P and a molecular mass near 334.22 g/mol. The beta anomer has a CAS Registry Number of 1094-61-7. It is typically supplied as a white to off-white powder for laboratory use. The molecule carries a phosphate group and a positively charged nicotinamide ring, giving it polar and water-soluble character. These properties influence how it is detected, purified, and stored in research and analytical laboratories.
Nicotinamide mononucleotide, abbreviated NMN, is a naturally occurring nucleotide. Its structure combines a nicotinamide ring, a ribose sugar, and a phosphate group. The compound exists in cells as an intermediate in the production of nicotinamide adenine dinucleotide, a central redox cofactor. NMN is distinct from nicotinamide riboside, another related pyridine nucleotide, although the two compounds can converge in metabolic pathways. Its chemical formula is C11H15N2O8P, and it carries a net negative charge at physiological pH.
In the salvage pathway, NMN is generated from nicotinamide and 5-phosphoribosyl-1-pyrophosphate by the enzyme nicotinamide phosphoribosyltransferase. A second route produces NMN from nicotinamide riboside through phosphorylation by nicotinamide riboside kinases. NMN is then converted to NAD+ by nicotinamide mononucleotide adenylyltransferases, often called NMNAT enzymes. This stepwise route allows cells to recycle nicotinamide and maintain NAD+ levels under changing metabolic conditions. The relative contribution of each route varies by tissue, species, and physiological state, and it remains an active area of research.
In the NAD+ salvage pathway, the enzyme NAMPT converts nicotinamide and a phosphate-donor molecule into NMN. A second enzyme, NMNAT, then converts NMN into NAD+. Nicotinamide riboside can also enter this route after being converted to NMN by nicotinamide riboside kinases. Because NMN sits at a junction between precursor uptake and NAD+ formation, its cellular concentration is tightly linked to enzyme activity and tissue type. NAD+ participates in redox reactions, signaling, and DNA repair, and its levels decline with age in some animal models, though human evidence remains more limited and context-dependent.
Research interest in NMN increased after animal studies reported that oral or injected NMN can raise NAD+ levels in some tissues. How NMN is absorbed and distributed in humans is not fully established. Some evidence suggests extracellular NMN may be dephosphorylated to nicotinamide riboside before cellular uptake, while other studies propose specific transport routes. Direct human data on these mechanisms remain limited. Regulatory status also varies: in some countries NMN is treated as a dietary supplement, while elsewhere it is restricted or requires approval, and these differences affect labeling, sale, and research.
=== Division of Israel and Judah === The monarchic state was divided into two states, Israel and Judah, due to civil and religious disputes. Eventually, Israel and Judah met their demise after the Assyrian and Babylonian invasions respectively. According to the Biblical prophets, these invasions were divine judgements for religious apostasy and corrupt leadership. This period is covered by 1 Kings 12 to 2 Kings 25 or alternatively, 2 Chronicles 10 to 2 Chronicles 36. The Book of Jonah narrates the prophet Jonah going to the Neo-Assyrian Empire to deliver a divine message.
== Contraindications and precautions == Patients with known hypersensitivity to Oprelvekin itself or any other ingredient. Patients with severe or decompensated heart failure should not be treated, because Oprelvekin may cause excessive fluid retention with edema and cardiac decompensation. Patients with compensated heart disease should be treated with caution and under permanent clinical supervision. Neumega is not indicated following myeloablative chemotherapy (increased likelihood of severe side-effects) and in pediatric patients. Renal impairment: Neumega is excreted renally. No differences of pharmakinetic parameters and clinical differences have been seen in mild to moderate impairment. Severe impairment has led to an increased number of patients with reduced hemoglobin due to dilutional anemia. Patients with severely disturbed renal function should be monitored very closely. The efficacy of Oprelvekin has not been systematically studied in patients receiving chemotherapy regimes of more than 5 days duration/each cycle or in those regimes containing agents that induce delayed thrombocytopenia (e.g. nitrosoureas, mitomycin C. Neumega should not be given in these cases.
=== Phylogenetic tree topologies and other parameters === Phylogenetic tree topologies are often the parameter of interest; thus, branch lengths and any other parameters describing the substitution process are often viewed as nuisance parameters. However, biologists are sometimes interested in the other aspects of the model. For example, branch lengths, especially when those branch lengths are combined with information from the fossil record and a model to estimate the timeframe for evolution. Other model parameters have been used to gain insights into various aspects of the process of evolution. The Ka/Ks ratio (also called ω in codon substitution models) is a parameter of interest in many studies. The Ka/Ks ratio can be used to examine the action of natural selection on protein-coding regions, it provides information about the relative rates of nucleotide substitutions that change amino acids (non-synonymous substitutions) to those that do not change the encoded amino acid (synonymous substitutions).

==== American involvement ==== In the mid-2010s, the US identified China as a primary source of illicit fentanyl and other synthetic opioids. American anti-drug intervention regarding China is influenced and constrained by the geopolitical sensitivities of US-China relations. The US employs several approaches: exerting diplomatic pressure on China to impose internal controls on illicit drugs; seeking cooperation in US and international law enforcement efforts; unilaterally imposing targeted sanctions; and bringing criminal indictments against Chinese companies and individuals. The effectiveness of these approaches depends significantly on China's level of implementation and enforcement, which is directly related to the fluctuating state of overall US-China relations. Following China's scheduling of fentanyl-class substances in 2019, the flow of synthetic opioids from China to the US appeared to subside; trafficking from China shifted to precursor chemicals and related equipment, and Mexico's drug cartels emerged as major clients. In November 2023, President Biden announced an agreement with General Secretary Xi Jinping for China to crack down on the export of precursor chemicals and pill presses to the Western Hemisphere. A US House committee report released in April 2024 found companies making fentanyl precursors in China can still apply for Chinese state tax rebates and other financial benefits after exporting the product.
Nuclear forensics is the investigation of nuclear materials to find evidence for the source, trafficking, and enrichment of the material. The material can be recovered from various sources including dust from the vicinity of a nuclear facility, or from the radioactive debris following a nuclear explosion. Results of nuclear forensic testing are used by different organisations to make decisions. The information is typically combined with other sources of information such as law enforcement and intelligence information.
